is a rare, potentially life-threatening autosomal recessive mechanobullous
disorder caused by mutations in genes encoding laminin-332 (LAMA3, LAMB3,
LAMC2) and collagen XVII (COL17A1), disrupting hemidesmosomal anchoring at the
lamina lucida level of the dermal-epidermal junction and producing
characteristic intra-lamina lucida cleavage upon minimal mechanical trauma.
Disease severity ranges from severe generalised JEB, formerly Herlitz type,
carrying high neonatal mortality from extensive blistering, sepsis, and
respiratory tract involvement, to milder generalised intermediate and localised
subtypes. Patients present with widespread fragile blistering, erosions,
granulation tissue formation around orifices, nail dystrophy, dental enamel
hypoplasia, and airway involvement in severe cases. Diagnosis integrates
