This gene encodes the proopiomelanocortin protein, expressed in skin components such as melanocytes, keratinocytes, and dermal microvascular endothelial cells. The inheritance pattern is autosomal recessive. Glucocorticoids, CRH, and ultraviolet radiation control POMC expression. It is a prohormone for adrenocorticotropic hormone (ACTH), a crucial player in the adrenocortical response to stress. Promoter methylation regulates POMC expression, while in ACTH-dependent Cushing's syndrome, the POMC gene is intrinsically activated. POMC consists of three segments: N-POMC (N-terminus), ACTH (middle), and β-LPH (C-terminus).
