BRAF mutations are rare in non-small cell lung cancer (NSCLC), occurring in about 1-5% of cases. However, they are most common in lung adenocarcinoma, a specific type of NSCLC. The most common BRAF mutation is V600E, found in more than 50% of cases with a BRAF mutation. V600E BRAF mutation is also the most common BRAF mutation in melanoma, a type of skin cancer. In melanoma, V600E BRAF mutation is associated with a significant dependency on MEK activity. This means cells with V600E BRAF mutation depend highly on the MEK protein to grow and survive. This provides a rational therapeutic strategy for V600E BRAF-mutant melanoma, as drugs that block MEK activity can effectively kill these cells. In lung adenocarcinoma, the frequency of V600E BRAF mutation is lower, at 1.5-2.8%.
