syndrome (retinal dystrophy, optic nerve edema, splenomegaly, anhidrosis, and
headache) is a rare autosomal dominant condition linked to heterozygous
mutations in the ALPK1 gene, which regulates innate immune
activation. The disease was initially described for its ocular manifestations,
including retinal degeneration, optic nerve swelling, and uveitis, but it is
now recognized to involve systemic features such as recurrent fever, arthritis,
splenomegaly, anhidrosis, and neurological changes including premature
