Hereditary
ATTR Amyloidosis Overview
Hereditary
ATTR Amyloidosis (hATTR) is a rare, autosomal dominant multisystem disorder
caused by mutations in the transthyretin (TTR) gene, which lead to misfolding
of the transthyretin protein and subsequent deposition of amyloid fibrils in
various tissues. These deposits disrupt normal organ function, most commonly
affecting the peripheral nerves, heart, gastrointestinal tract, and kidneys,
though the central nervous system and eyes may also be involved in certain
variants. Clinically, hATTR presents with a spectrum of symptoms depending on
the mutation and organ involvement—ranging from progressive sensorimotor
polyneuropathy and autonomic dysfunction to restrictive cardiomyopathy,
arrhythmias, and heart failure. The disease often manifests in adulthood, with
variable penetrance and age of onset, and may be misdiagnosed due to its
overlap with other neuropathic or cardiac conditions.
