Dyskeratosis
Congenita (DC) Overview
Dyskeratosis
Congenita (DC) is a rare genetic syndrome characterized by nail, skin, and oral
issues, along with a heightened risk of bone marrow failure and cancer. It
results from mutations in various genes linked to telomere biology. DKC1
mutations cause X-linked recessive DC, while autosomal dominant genes involved
are TERC, TERT, TINF2, RTEL1, PARN, and ACD. Autosomal recessive genes include
TERT, NHP2, NOP10, RTEL1, WRAP53, and ACD, with CTC1, POT1, and STN1 as rare
causes. DC presents with nail abnormalities, skin changes, and oral leukoplakia
in childhood and may lead to bone marrow failure and cancer. Diagnosis involves
telomere length testing, and management includes regular check-ups and, in the
case of bone marrow failure, hematopoietic stem cell transplantation, or
androgen therapy. Life expectancy varies, with a significant risk of bone
marrow failure, cancer, and pulmonary complications.
