LAMA2-Related Dystrophy (LAMA2-RD)
Overview
LAMA2-Related
Dystrophy (LAMA2-RD) is a genetic disorder that primarily affects skeletal
muscles, leading to weakness and wasting. It is caused by mutations in the
LAMA2 gene, which is responsible for producing laminin-alpha2, a protein
crucial for the stability and function of muscle cells. The severity of
LAMA2-RD varies widely, ranging from severe early-onset forms to milder
late-onset presentations. In its severe form, LAMA2-RD manifests at birth or
within the first few months of life. Infants with this condition often exhibit
profound muscle weakness, low muscle tone (hypotonia), and limited spontaneous
movement. Feeding difficulties, respiratory insufficiency, and joint
deformities are common complications. As affected children grow, they may
develop scoliosis and other spinal abnormalities, and most do not achieve
independent walking. Seizures and, in rare cases, heart complications may also
occur.
