Systemic
mastocytosis is a diverse disorder marked by the accumulation of malignant mast
cells in the bone marrow and various organs. The precise cause is not fully
understood, although an activating mutation of the KIT gene, typically D816V,

lineages. In advanced systemic mastocytosis (AdvSM), additional mutations in
genes like TET2, SRSF2, ASXL1, RUNX1, JAK2, N/KRAS, CBL, and less frequently,
EZH2, IDH2, ETV6, U2AF, or SF3B1, contribute to disease aggressiveness,
especially in multi-mutated cases. Over 80% of systemic mastocytosis patients

indolent disease, a subset faces aggressive variants collectively termed
“advanced systemic mastocytosis,” including aggressive systemic mastocytosis,
