Essential Thrombocythemia – Market Outlook, Epidemiology, Competitive Landscape, and Market Forecast Report – 2023 To 2033
Essential thrombocytosis (ET), also known as essential thrombocythemia, is a myeloproliferative neoplasm caused by the overproduction of hematopoietic cells due to mutations in the JAK2, CALR, or MPL genes. These genes are called "driver mutations" due to their crucial role in developing myeloproliferative neoplasms. The JAK2 gene, in particular, has a specific point mutation known as JAK2V617F that leads to a gain of function and activation of intracellular signaling pathways involved in hematopoietic cytokines. Half of ET patients have JAK2 mutations, while CALR mutations result from insertions or deletions causing a shift in the amino acid reading frame. MPL mutations occur through point mutations in about 3 to 15% of ET patients. Hydroxyurea is the first-line cytoreductive therapy for ET, while anagrelide is used as a second-line therapy if hydroxyurea is not well-tolerated or not effective. Hydroxyurea reduces platelet and leukocyte count, thus reducing the risk of thrombosis and myelofibrosis. Anagrelide, on the other hand, is specifically used to reduce platelet count.
- The annual incidence of Essential thrombocytosis in France ranges from 0.59 to 2.53 per 100,000.
