Alzheimer's disease (AD) is a progressive neurodegenerative disease associated with cognitive decline and is the most common form of dementia in the elderly. Early-onset familial AD, which typically develops before the age of 65 years and accounts for only a small portion (<1%) AD cases, is primarily caused by overproduction of Aβ owing to mutations in either the APP gene or genes encoding presenilin 1 (PSEN1) or presenilin 2 (PSEN2), essential components of the γ-secretase complexes responsible for cleavage and release of Aβ.

·       The ε4 allele of the apolipoprotein E (APOE) gene is the strongest risk factor for LOAD.

·       The human APOE gene exists as three polymorphic alleles ε2, ε3, and ε4, which have a worldwide frequency of 8.4%, 77.9%, and 13.7%, respectively. However, the frequency of the ε4 allele is dramatically increased to ~40% in patients with AD.
