Multiple myeloma (MM) is a hematologic malignancy characterized by the uncontrolled proliferation of clonal plasma cells in the bone marrow, which secrete significant quantities of non-functional proteins and immunoglobulins. The primary objective in treating relapsed MM is to alleviate disease symptoms and thwart the emergence of CRAB symptoms, defined as elevated calcium levels (>11.5 mg/dL), increased creatinine levels (>2 mg/dL), low hemoglobin levels (<10 g/dL or >2 points below the lower normal limit), and the development of new bone lesions. Managing relapsed and refractory MM involves a multifaceted treatment approach, which includes a standard combination of proteasome inhibitors, immunomodulatory drugs, and monoclonal antibodies. Additionally, treatment may incorporate histone deacetylase inhibitors and alkylating agents.
