For diagnostic purposes, the Fluorescence in situ hybridization (FISH) assay, particularly the Vysis® ALK Break-Apart FISH Probe Kit test from Abbott Molecular, stands as the recommended method for detection. Various second and third-generation tyrosine kinase inhibitors (TKIs) have been formulated to address resistance to treatment, effectively targeting numerous ALK resistance mutations. Notably, studies have shown that first-line therapy employing alectinib, ceritinib, lorlatinib, or brigatinib surpasses crizotinib or chemotherapy, where applicable, in managing patients with ALK-rearranged disease.
