Mantle cell lymphoma (MCL) is caused by the malignant transformation of a B lymphocyte on the rim of a lymph node follicle (the mantle zone). The transformed B lymphocyte grows uncontrollably, resulting in the accumulation of lymphoma cells, which causes the enlargement of lymph nodes. MCL cells can enter lymphatic channels and the blood, spreading to other lymph nodes or tissues like the bone marrow, liver, and gastrointestinal tract. In almost all cases, MCL is distinguished by the overexpression of cyclin D1 (a protein that promotes cell growth). Cyclin D1 overexpression is typically caused by a rearrangement (translocation) between chromosomes 11 and 14.
